Background
IgG4 is a subclass of immunoglobulin G (IgG), which comprises four main subclasses: IgG1, IgG2, IgG3, and IgG4. IgG4 primarily participates in immune regulation within the immune system and plays a significant role in certain inflammatory responses. For instance, it can form cross-links with other antibodies, preventing them from binding to antigens, thereby reducing the intensity of inflammatory reactions. Elevated IgG4 levels are often observed in autoimmune diseases such as systemic lupus erythematosus, rheumatoid arthritis, and Sjögren's syndrome.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human tonsil. Anti-Human IgG4 antibody was used at 1/2000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
