Background

Insulin-like growth factor-binding protein 2 (IGFBP2) is a member of the IGFBP family, which regulates the bioavailability and activity of insulin-like growth factors (IGF-I and IGF-II). Primarily secreted by the liver, IGFBP2 modulates cell proliferation, differentiation, and metabolism by binding IGFs with high affinity, preventing their interaction with IGF receptors. Beyond its IGF-dependent roles, IGFBP2 exhibits IGF-independent functions, influencing processes like apoptosis, angiogenesis, and tumor progression. Elevated IGFBP2 levels are associated with various cancers, including glioblastoma and prostate cancer, where it may promote invasiveness. Conversely, IGFBP2 also plays a role in metabolic disorders, such as obesity and diabetes, highlighting its diverse physiological and pathological significance.
