Background
FcRn is a critical protein responsible for regulating the lifespan and transport of immunoglobulin G (IgG) antibodies. Primarily located in endothelial and phagocytic cells, FcRn binds to IgG in acidic endosomes, recycling them back to the cell surface and releasing them into circulation at a neutral pH. This unique pH-dependent binding mechanism protects IgG from lysosomal degradation, significantly extending its serum half-life. Additionally, FcRn mediates the transcytosis of IgG across placental and epithelial barriers, facilitating passive immunity from mother to offspring and mucosal immune defense. Its role makes it a major therapeutic target for modulating antibody-based therapies.
