Background
The Epidermal Growth Factor Receptor (EGFR) is a transmembrane tyrosine kinase receptor belonging to the ErbB family. Upon binding to its ligands such as EGF or TGF-α, EGFR undergoes homo- or hetero-dimerization, triggering autophosphorylation of intracellular tyrosine residues and activation of downstream signaling cascades including MAPK/ERK, PI3K/AKT, and JAK/STAT pathways. These regulate critical cellular processes including proliferation, survival, migration, and differentiation. EGFR is frequently overexpressed, amplified, or mutated in various cancers, particularly non-small cell lung cancer (NSCLC), colorectal cancer, and glioblastoma.
