Background

Ribonuclease 3, commonly known as Drosha, is a Class 2 ribonuclease III enzyme crucial for microRNA (miRNA) biogenesis. Located in the nucleus, it functions as the core catalytic component of the Microprocessor complex. Its primary role is to initiate miRNA processing by cleaving long primary miRNA transcripts (pri-miRNAs) into approximately 70-nucleotide precursor miRNAs (pre-miRNAs). This precise endonucleolytic cleavage is the first and essential step in the miRNA maturation pathway, which ultimately regulates gene expression post-transcriptionally. Dysregulation of Drosha is implicated in various cancers and developmental disorders.
