Background
Human Coagulation Factor XI (FXI) is a plasma glycoprotein synthesized primarily in the liver and is a key zymogen within the intrinsic coagulation pathway. Upon activation by Factor XIIa, thrombin, or auto-activation, it converts to the serine protease Factor XIa. This active form then specifically cleaves and activates Factor IX, thereby amplifying the downstream coagulation cascade to generate thrombin and form a stable fibrin clot. Beyond its procoagulant role, FXI is also implicated in various thrombo-inflammatory processes. Notably, genetic deficiency of FXI results in a mild bleeding tendency known as hemophilia C, while elevated levels are considered a risk factor for thrombotic diseases like stroke and venous thromboembolism. Consequently, Factor XI is now a prominent therapeutic target, with inhibitors under development to create safer anticoagulants with potentially reduced bleeding risks.

Sample is undiluted samples are as follows: HepG2 cell culture supernatant (50%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 2.96 ng/mL in HepG2 cell culture supernatant.
