Background
Complement Factor B is a pivotal zymogen in the alternative pathway of the complement system. This serine protease circulates in plasma and is essential for innate immunity. Its activation occurs when it binds to C3b, forming the C3 convertase (C3bBb) upon cleavage by Factor D. This enzyme complex amplifies the complement response by cleaving more C3, driving opsonization, inflammation, and the formation of the membrane attack complex. Factor B also plays a role in the lectin pathway. As a key regulator, its deficiency or dysfunction is linked to susceptibility to infections and autoimmune diseases like atypical hemolytic uremic syndrome.
