Background

HTRA1 (High Temperature Requirement A Serine Peptidase 1) is a secreted serine protease belonging to the HtrA family, primarily known for its critical role in regulating extracellular matrix (ECM) homeostasis through the degradation of key structural proteins such as fibronectin, vitronectin, and various small leucine-rich proteoglycans. Beyond its proteolytic function, HTRA1 acts as a potent antagonist of the TGF-β signaling pathway by cleaving TGF-β binding proteins, thereby modulating cell proliferation, differentiation, and tissue repair mechanisms. Dysregulation of HTRA1 activity has been strongly implicated in several pathological conditions, most notably age-related macular degeneration (AMD), where gain-of-function mutations lead to excessive ECM breakdown in the retina, as well as in certain cancers and connective tissue disorders like CARASIL (Cerebral Autosomal Recessive Arteriopathy with Subcortical Infarcts and Leukoencephalopathy), highlighting its significance as both a diagnostic biomarker and a potential therapeutic target in maintaining tissue integrity and preventing degenerative diseases.
