Background

HTR2A, also known as the 5-hydroxytryptamine receptor 2A, is a G protein-coupled receptor composed of 471 amino acids with a molecular weight of approximately 52 kDa. It serves as a key receptor for the neurotransmitter serotonin (5-hydroxytryptamine) in the brain and is widely distributed in the cerebral cortex and basal ganglia, as well as in smooth muscle cells of peripheral tissues. When activated by serotonin or psychoactive substances such as LSD and psilocybin, HTR2A primarily couples to the Gq protein, activating phospholipase C and triggering a cascade of intracellular calcium release and downstream signaling pathways, thereby modulating neural activity, perception, cognition, and emotion. Consequently, dysfunction of HTR2A is closely associated with various psychiatric disorders, including schizophrenia, depression, and substance dependence, making it an important therapeutic target for the development of antipsychotic and antidepressant drugs.

Immunocytochemistry

ICC shows positive staining in U-87 MG cells. Anti-HTR2A antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
