Background
HSP90 (Heat Shock Protein 90) is a highly conserved and extremely abundant molecular chaperone protein within cells. Existing as a homodimer, it assists numerous "client proteins" in proper folding, maturation, and activity maintenance through ATP-dependent conformational changes. These client proteins are predominantly kinases, hormone receptors, and transcription factors involved in cell proliferation, survival, and signal transduction. Its expression is upregulated when cells respond to external stresses such as heat shock and oxidative stress, where it exerts important cytoprotective functions by stabilizing denatured proteins and preventing their aggregation. Additionally, HSP90 participates in immune regulation—for example, by promoting antigen presentation and TLR signaling pathway activation to enhance innate immune responses, as well as supporting T cell proliferation and differentiation. Since its client protein network includes a large number of oncoproteins (such as HER2, AKT, and BRAF), HSP90 inhibition has become an important strategy in cancer therapy. Furthermore, its dysfunction is closely associated with the accumulation of protein aggregates in neurodegenerative diseases (such as Alzheimer's disease) as well as with fungal infection drug resistance.
