Background
Heat shock factor 1 (HSF1) is an evolutionarily conserved transcription factor that acts as the master regulator of the inducible heat-shock response by binding to heat-shock elements (HSEs) in the promoters of cytoprotective chaperone genes such as HSP70 and HSP90, thereby driving rapid expression of molecular chaperones that prevent stress-induced protein misfolding and aggregation; beyond this canonical cytoprotective role, HSF1 also modulates diverse cellular processes including development, metabolism, apoptosis and aging, is frequently hijacked by cancers to sustain oncogenic protein homeostasis and promote malignant proliferation, and is therefore emerging as a key therapeutic target in both protein-misfolding disorders and cancer.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human colon. Anti-HSF1 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry
ICC shows positive staining in HeLa cells. Anti-HSF1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
