Background

HIRA (Histone Cell Cycle Regulator) is a conserved histone chaperone that forms a multi-subunit complex—typically including UBN1, CABIN1, and ASF1a—to deposit the histone variant H3.3 into chromatin independently of DNA replication, thereby modulating nucleosome dynamics at transcriptionally active promoters, enhancers, and gene bodies; by stabilizing open chromatin states, HIRA underlies gene activation, embryonic development, stress-responsive transcription, senescence-associated heterochromatin formation, and DNA-damage repair, while its dysregulation or haploinsufficiency at human chromosome 22q11.21 has been linked to DiGeorge syndrome and tumorigenesis.
