Background

Helz2 (Helicase with Zinc Finger 2) is a large, multifunctional nuclear protein belonging to the DEAH-box family of RNA helicases, characterized by its extensive domain architecture that includes an N-terminal helicase core, multiple zinc finger motifs, and a C-terminal region involved in protein-protein interactions. Primarily recognized for its critical role as a transcriptional coactivator for the thyroid hormone receptor (TR), Helz2 facilitates gene expression by recruiting other cofactors and modifying chromatin structure in a ligand-dependent manner, thereby influencing metabolic regulation, development, and cellular differentiation. Beyond its endocrine functions, recent studies have implicated Helz2 in various other biological processes, including the regulation of circadian rhythms, DNA damage response, and the modulation of signaling pathways such as Wnt/β-catenin, while genetic variations in the HELZ2 locus have been associated with susceptibility to certain cancers and metabolic disorders, highlighting its significance as a potential therapeutic target and biomarker in human disease.
