Background

HDAC3 (histone deacetylase 3) is a 428-amino-acid zinc-dependent enzyme that removes acetyl groups from lysine residues on histones and many non-histone transcription factors, thereby compacting chromatin and repressing gene expression; it is an obligate component of the nuclear receptor corepressor (NCoR/SMRT) complex, shuttles between nucleus and cytoplasm through an intrinsic nuclear export signal, and its catalytic activity is allosterically activated by inositol tetraphosphate (IP4) bound within a conserved pocket, while global deletion in mice is embryonic-lethal and tissue-specific deletion reveals essential roles in circadian rhythm, immune tolerance, DNA repair, lipid and glucose metabolism, and neuronal function, making HDAC3 a major epigenetic drug target for cancer, metabolic and inflammatory diseases.
