Background

Phospho-GSK3α/β (Ser21/9) is the phosphorylated, inactive form of glycogen synthase kinase-3 alpha and beta isoforms, generated when AKT, MAPK and other kinases add a phosphate to Ser21 of GSK3α or the homologous Ser9 of GSK3β, thereby creating an N-terminal pseudo-substrate that folds back into the catalytic pocket to block substrate access and halt the enzyme’s constitutive suppression of glycogen synthesis, β-catenin signaling, NF-κB activity and other metabolic/transcriptional programs; detection of this phospho-epitope therefore serves as a widely used biomarker for PI3K–AKT–mTOR pathway activation and for cellular responses to insulin, growth factors, oxidative stress and Wnt stimuli.
