Background

GRB2 (growth factor receptor-bound protein 2) is a widely expressed adaptor protein that plays a central bridging role in multiple cellular signal transduction pathways. Its structure is highly conserved, consisting of one SH2 domain flanked by two SH3 domains on either side. The SH2 domain recognizes and binds to phosphorylated tyrosine residues, such as those found on activated receptor tyrosine kinases (e.g., EGFR and PDGFR), while the SH3 domains interact with downstream effector molecules, such as SOS (a guanine nucleotide exchange factor), by binding to proline-rich motifs. Through this modular structure, GRB2 connects activated receptors to the Ras-MAPK signaling pathway: upon growth factor stimulation, GRB2 binds via its SH2 domain to the phosphorylated receptor and subsequently recruits SOS to the plasma membrane through its SH3 domains, promoting the conversion of Ras from its inactive GDP-bound state (Ras-GDP) to its active GTP-bound state (Ras-GTP), ultimately initiating the MAPK/ERK cascade to regulate cell proliferation, differentiation, and survival. In addition to the classical Ras pathway, GRB2 is also involved in the regulation of PI3K-AKT, JNK, and FAK signaling pathways, and plays roles in immune cell activation, cytoskeletal remodeling, and cell migration. GRB2 is overexpressed in various tumors and is considered a promoter of tumor initiation and progression. Particularly in breast cancer, liver cancer, and pancreatic cancer, its aberrant expression is closely associated with tumor malignancy and poor prognosis, making it a potential intervention target in cancer therapeutic research. Moreover, mutations in GRB2 in human genetic disorders are also linked to certain immunodeficiencies and developmental abnormalities, further underscoring its essential role in normal physiology.

Immunohistochemistry

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
