Background

GPX4 (Glutathione Peroxidase 4) is a pivotal antioxidant enzyme, unique for its active site containing the rare amino acid selenocysteine, which is essential for its function. It directly reduces a broad spectrum of phospholipid hydroperoxides within cell membranes, thereby maintaining membrane integrity and playing a central role in inhibiting ferroptosis—a form of iron-dependent, non-apoptotic cell death. Its functions are critical for sperm development, T-cell immunity, and neuronal survival. Clinically, GPX4 is a key therapeutic target, as its dysfunction is implicated in conditions ranging from male infertility and neurodegenerative diseases to epilepsy and cancer, with modulating its activity being a promising strategy for treating ferroptosis-related pathologies.
