Background
GPR75, or G protein-coupled receptor 75, is an orphan G protein-coupled receptor primarily expressed in the brain, particularly within the hypothalamus, where it plays a critical role in regulating energy homeostasis and body weight. Recent genetic studies have identified GPR75 as a significant protective factor against obesity, demonstrating that loss-of-function mutations or knockout of this receptor in mice leads to resistance to diet-induced obesity, improved insulin sensitivity, and increased energy expenditure without affecting food intake. Mechanistically, GPR75 is believed to interact with signaling pathways involving the melanocortin system and may respond to specific lipid ligands, thereby influencing neuronal activity in appetite-regulating centers.
