Background

GPR3 (G protein-coupled receptor 3) is an orphan GPCR primarily expressed in the mammalian brain and oocytes, belonging to the Class A family. It is characterized by exceptionally high constitutive Gs protein activity, continuously activating adenylyl cyclase and elevating intracellular cAMP levels in the absence of exogenous ligands. Functionally, GPR3 promotes neuronal synapse growth, axon polarization, and anti-apoptosis through signaling pathways such as cAMP/PKA and PI3K/Akt, exerting important neuroprotective effects. Meanwhile, it maintains meiotic arrest in oocytes until the luteinizing hormone surge occurs. Recent studies have identified oleoylethanolamide (OEA) as its endogenous agonist, elucidated its cryo-EM structure in complex with G proteins, and revealed a regulatory mechanism involving dimerization-mediated autoinhibition. Dysfunction of GPR3 is closely associated with Alzheimer's disease (by modulating γ-secretase to promote Aβ generation), neuropathic pain, and premature ovarian failure, making it an important potential drug target for various diseases.
