Background

GPHN (gephyrin) is a 93-kDa multifunctional scaffold protein that self-assembles into a hexagonal lattice beneath the postsynaptic membrane of inhibitory synapses, where it simultaneously anchors both glycine and GABA type A receptors to subsynaptic microtubules and actin cytoskeleton, thereby clustering and immobilizing these receptors to ensure reliable inhibitory neurotransmission, while also possessing dual enzymatic activities (molybdopterin adenylyltransferase and molybdotransferase) essential for the biosynthesis of the molybdenum cofactor, and its alternative splicing, extensive post-translational modifications and dynamic exchange at synapses modulate inhibitory synaptic strength and plasticity, with mutations or altered expression of GPHN linked to epilepsy, autism spectrum disorders, schizophrenia and molybdenum cofactor deficiency.
