Background

Glypican-3 (GPC3) is a cell-surface glycoprotein that is part of the heparan sulfate proteoglycan family. It is primarily expressed in the placenta and fetal liver, with limited expression in adult tissues, making it a promising biomarker for certain cancers. GPC3 is overexpressed in hepatocellular carcinoma (HCC) and other cancers such as ovarian clear cell carcinoma, melanoma, and pediatric cancers like hepatoblastoma and Wilms' tumor. This protein plays a crucial role in various cellular processes, including cell proliferation, migration, and differentiation, by interacting with growth factors such as Wnt, fibroblast growth factor (FGF), and insulin-like growth factor (IGF). Its re-expression in HCC and its involvement in tumor-associated signaling pathways make it a valuable target for both diagnostic and therapeutic strategies. GPC3 has been used as an immunohistochemical marker for HCC diagnosis, and its presence in serum can serve as a liquid biopsy marker. Additionally, various immunotherapies targeting GPC3, including monoclonal antibodies and CAR-T cell therapies, are being developed to inhibit tumor growth and improve clinical outcomes.
