Background

The fourth gene of the Ebola virus (EBOV) encodes the glycoprotein GP, a multidomain and multifunctional class I fusion protein. It exists as trimers on the viral surface and mediates viral entry through a heterodimer of GP1 and GP2 subunits generated by furin cleavage. GP1 facilitates infection via its receptor-binding domain (RBD), which interacts with key host receptors such as TIM-1 and NPC1, while its mucin-like domain—modified by glycosylation—enhances infectivity and shields neutralization epitopes. GP2 drives membrane fusion and nucleocapsid release. Additionally, GP exhibits significant immunomodulatory functions: it downregulates integrin expression to disrupt vascular integrity, activates monocytes/macrophages via the TLR4 pathway triggering a cytokine storm, counteracts the antiviral effects of BST2, and acts as a decoy to evade antibody neutralization. These mechanisms collectively promote immune evasion and contribute to viral pathogenesis.
