Background

GLP1R (Glucagon-like peptide-1 receptor) is a G protein-coupled receptor (GPCR) primarily expressed in pancreatic β-cells, as well as in the brain, heart, and gastrointestinal tract. It is activated by the incretin hormone GLP-1 (glucagon-like peptide-1), which is secreted by intestinal L-cells in response to nutrient ingestion. Upon activation, GLP1R stimulates glucose-dependent insulin secretion, suppresses glucagon release, delays gastric emptying, and promotes satiety—making it a critical therapeutic target for type 2 diabetes mellitus and obesity. Notably, GLP1R agonists such as semaglutide and liraglutide have revolutionized metabolic disease management by mimicking endogenous GLP-1 action with enhanced stability and potency. Additionally, emerging evidence suggests neuroprotective and cardioprotective roles of GLP1R signaling, expanding its clinical relevance beyond glycemic control.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human pancreas. Anti-GLP1R antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Negative control: IHC shows negative staining in paraffin-embedded human tonsil. Anti-GLP1R antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
