Background

GDNF family receptor alpha-1 (GFRA1) is a 465-amino-acid, glycosylphosphatidylinositol-anchored cell-surface glycoprotein that functions as the high-affinity ligand-binding component for glial-cell-line-derived neurotrophic factor (GDNF) and its close homolog neurturin; upon ligand binding GFRA1 nucleates formation of a tripartite complex that recruits and trans-activates the transmembrane tyrosine kinase RET, thereby triggering downstream survival, proliferation and neurite-outgrowth signals (PI3K/AKT, RAS/MAPK, PLCγ) in dopaminergic, motor, sensory and enteric neurons, and this same GFRA1-mediated signaling is essential during kidney morphogenesis, spermatogenesis and peripheral-nerve repair while also being implicated in cancer chemoresistance and Parkinson’s disease.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-GDNF Receptor alpha 1/GFRA1 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
