Background

GCLM (glutamate-cysteine ligase modifier subunit) is the regulatory component of glutamate-cysteine ligase (GCL), the rate-limiting enzyme in glutathione biosynthesis; by forming a heterodimer with the catalytic GCLC subunit, GCLM increases the enzyme’s affinity for glutamate and ATP, raises its Ki for glutathione feedback inhibition, and thus accelerates production of the antioxidant glutathione that maintains cellular redox balance and protects against oxidative stress.

Immunohistochemistry

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
