Background
GCC1, also known as GRIP and coiled-coil domain-containing protein 1 or GCC185, is a large peripheral membrane protein localized primarily to the trans-Golgi network (TGN) that functions as an essential tethering factor in retrograde transport by anchoring Rab9-positive vesicles carrying mannose-6-phosphate receptors from late endosomes back to the TGN. Structurally characterized by extensive coiled-coil domains and multiple Rab-binding sites, GCC1 interacts not only with active Rab9 GTPase but also with SNARE proteins such as syntaxin 6 and syntaxin 16, thereby facilitating vesicle docking and fusion while maintaining Golgi architecture and ensuring proper lysosomal enzyme sorting; consequently, dysfunction or depletion of GCC1 disrupts endosome-to-TGN trafficking, leading to missorting of cargo, fragmented Golgi morphology, and impaired cellular homeostasis, highlighting its critical role in intracellular logistics and organelle integrity.
