Background
Guanylate-binding protein 2 (GBP2) is a core component of the innate immune system, induced by interferons (IFNs) and belonging to the dynamin-related GTPase family. It binds and hydrolyzes GTP, with GDP being its primary hydrolysis product. Functionally, GBP2 serves as a critical effector molecule against a broad spectrum of bacterial, viral, and protozoan pathogens. Its mechanisms of action include: on one hand, it can rupture pathogen-containing vacuoles, releasing pathogens into the cytosol and thereby activating the inflammasome signaling pathway to trigger robust immune responses; on the other hand, it can exert antiviral effects independently of its GTPase activity by inhibiting the maturation of viral envelope proteins. Structurally, GBP2 possesses a globular G domain and a flexible C-terminal helical domain. Its GTPase activity is tightly regulated, and it undergoes dynamic interconversion between monomeric and dimeric states during the GTP hydrolysis cycle. These sophisticated structural and functional features position GBP2 as a key player in cell-autonomous immunity.

IHC shows positive staining in paraffin-embedded human spleen. Anti-GBP2 antibody was used at 1/400 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
