Background

GBA protein, encoded by the GBA gene on chromosome 1q21, is a 497-amino-acid lysosomal glucocerebrosidase that catalyzes the hydrolysis of glucosylceramide to glucose and ceramide; homozygous mutations cause Gaucher disease, while heterozygous mutations are the strongest common genetic risk factor for Parkinson’s disease and dementia with Lewy bodies through gain-of-function mechanisms that promote α-synuclein aggregation, impaired autophagy-lysosomal flux, and neuroinflammation, and its enzymatic activity can be enhanced by small-molecule chaperones or gene therapy, making it a key therapeutic target for both systemic and neurodegenerative disorders.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human colon. Anti-GBA antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
