Background
Gamma Cystathionase, also known as cystathionine γ-lyase (CSE), is a multifunctional pyridoxal phosphate-dependent enzyme encoded by the CTH gene. It plays a central role in the transsulfuration pathway, catalyzing the final step in the conversion of methionine to cysteine, namely the cleavage of cystathionine into cysteine, α-ketobutyrate, and ammonia. Beyond this classical function, the enzyme exhibits broad substrate specificity and can utilize substrates such as cysteine and homocysteine to generate the important gaseous signaling molecule hydrogen sulfide (H₂S), thereby participating in various physiological processes including blood pressure regulation, angiogenesis, and resistance to oxidative stress. It can also regulate the function of target proteins (such as GAPDH and NF-κB) through a post-translational modification known as "persulfidation." In humans, deficiency of this enzyme leads to cystathioninuria, an autosomal recessive disorder characterized by abnormal accumulation of cystathionine.
