Background

GABA-A receptor alpha 5 (GABRA5) is a subunit of the type-A gamma-aminobutyric acid receptor, a ligand-gated chloride channel that mediates the major form of fast inhibitory neurotransmission in the mammalian brain. The alpha5 subunit co-assembles with beta (beta1 or beta3) and gamma2 subunits to form heteropentameric receptors, which are predominantly expressed in the hippocampus and localized largely extrasynaptically on pyramidal cell dendrites. These receptors exhibit high sensitivity to ambient GABA and slow desensitization, making them well-suited for mediating tonic inhibition, a persistent form of inhibitory conductance that regulates neuronal excitability. Functionally, alpha5-containing GABA-A receptors play a critical role in learning and memory processes, and studies strongly suggest that amnesia is primarily mediated by alpha5-beta-gamma2 receptors. Genetic variations in GABRA5 have been linked to susceptibility to panic disorder and bipolar affective disorder, and de novo mutations in the GABRA5 gene cause developmental and epileptic encephalopathy-79 (DEE79), characterized by severe seizures and impaired cognitive development. Additionally, the alpha5 subunit is a promising pharmacological target, with inverse agonists showing potential for promoting recovery after stroke by reducing excessive tonic inhibition in the peri-infarct zone.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
