Background

FOXP4 (Forkhead box protein P4) is a functionally critical transcription factor belonging to the FOXP subfamily, predominantly exerting transcriptional repressive functions within the nucleus. Its activity depends on forming homodimers via its leucine zipper domain or forming heterodimers with FOXP1 and FOXP2 to bind DNA. In physiological development, it is a multifunctional regulator essential for the morphogenesis of organs such as the heart, lung, and intestine, playing critical roles in embryonic foregut morphogenesis, pulmonary secretory cell differentiation, and the maintenance of cerebellar Purkinje cell dendrites. However, dysregulation of FOXP4 is closely associated with various diseases: heterozygous mutations or homozygous loss-of-function variants have been confirmed to cause autosomal dominant or recessive genetic disorders, with patients typically presenting with short stature, developmental malformations, neurodevelopmental disorders, congenital heart disease, and language developmental delay among a spectrum of congenital abnormalities. Additionally, FOXP4 plays a complex "dual-faceted" role in tumorigenesis—it may act as a tumor suppressor downregulated in renal cancer, but conversely functions as an oncogene overexpressed in non-small cell lung cancer, hepatocellular carcinoma, and prostate cancer, where it promotes tumor growth and invasion.

The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
