Background

FOXO4 (Forkhead Box O4) is a transcription factor belonging to the FOXO family of proteins, which function as critical regulators of cellular stress responses, metabolism, longevity, and apoptosis. As a key downstream effector of the PI3K-AKT signaling pathway, FOXO4 activity is tightly controlled by post-translational modifications, particularly phosphorylation by AKT, which promotes its nuclear exclusion and cytoplasmic retention, thereby inhibiting its transcriptional functions. Upon activation by various stress stimuli—including oxidative stress, DNA damage, and nutrient deprivation—FOXO4 translocates to the nucleus where it binds to specific DNA sequences (FKHR recognition motifs) to modulate the expression of target genes involved in cell cycle arrest (p21, GADD45), DNA repair, autophagy, and pro-apoptotic pathways (BIM, FASL). Notably, FOXO4 has garnered significant attention in aging research due to its role in cellular senescence; the FOXO4-DRI (D-Retro-Inverso) peptide, which disrupts the interaction between FOXO4 and p53, has been demonstrated to selectively induce apoptosis in senescent cells, thereby extending healthspan and lifespan in aged mice. Additionally, FOXO4 participates in diverse physiological processes including glucose homeostasis, adipogenesis, and stem cell maintenance, while its dysregulation has been implicated in various pathologies such as cancer, where it can function as either a tumor suppressor or promoter depending on cellular context, as well as in neurodegenerative diseases and metabolic disorders.

Immunocytochemistry

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
