Flow cytometric analysis of Human CD63 expression on human PBMC (human peripheral blood mononuclear cell). Human PBMC were stained with PE-Cy7 Mouse Anti-Human CD123 Antibody and either FITC Mouse IgG1, k Isotype Control (left panel) or SDT FITC Mouse Anti-Human CD63 Antibody (right panel) at 5 μl/test. Total viable cells, as determined by Fixable Viability Dye 452 (S0D0021), were used for analysis. Flow cytometry and data analysis were performed using Agilent NovoCyte Quanteon and FlowJo™ software.

Background
CD63 is a transmembrane glycoprotein belonging to the tetraspanin superfamily, characterized by four transmembrane domains, a small extracellular loop, and a large extracellular loop (EC2) that often undergoes N-glycosylation. It is widely expressed in various cell types (such as platelets, immune cells, endothelial cells, and cancer cells), and is particularly enriched on the membranes of activated platelets and late endosomes/lysosomes. As a late endosomal/lysosomal membrane protein, CD63 serves as a classic marker of extracellular vesicles (especially exosomes) and is commonly used to identify and isolate exosomes. Functionally, CD63 is involved in regulating intracellular vesicle trafficking, protein sorting, cell adhesion, migration, and signal transduction, and also mediates pathogen invasion. Furthermore, its abnormal expression is associated with various pathological processes—for example, it is highly expressed in cancers such as melanoma and lung cancer, promoting tumor invasion and metastasis. It also undergoes external translocation from lysosomal membranes during platelet activation and plays important roles in allergic diseases, viral infections, and neurodegenerative disorders. Therefore, CD63 is widely used as a biomarker for exosome tracking, disease diagnosis, and targeted therapy.
