Flow cytometric analysis of human CD319 expression on human PBMC. Human PBMC were stained with Brilliant Violet 421™ Mouse Anti-Human CD3 Antibody and either FITC Mouse IgG2b, k Isotype Control (left panel) or SDT FITC Mouse Anti-Human CD319 Antibody (right panel) at 5 μl/test. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
CD319, also termed SLAMF7 or CS1, is a 66 kDa self-ligating type-I trans-membrane glycoprotein of the CD2/SLAM immunoglobulin superfamily that is constitutively expressed on NK cells, activated B cells, monocytes and, at very high levels, on multiple-myeloma cells; its cytoplasmic tail contains ITSM sequences that, depending on the simultaneous presence or absence of the adaptor proteins EAT-2 and SAP, can switch between activating (via PI3K-PLCγ-ERK) and inhibitory signaling, thereby modulating NK-cell cytotoxicity, B-cell proliferation, macrophage cytokine release and antibody-dependent myeloma killing, making it both an immunomodulatory receptor and the therapeutic target of the FDA-approved monoclonal antibody elotuzumab.
