Background

Human Epidermal growth factor Receptor 2 (HER2) is also called ERBB2, HER-2, NEU, NGL, TKR1 and c-erb B2, and is a protein giving higher aggressiveness in breast cancers. It is a member of the ErbB protein family, more commonly known as the epidermal growth factor receptor family. HER2 protein cannot bind growth factors due to the lacking of ligand binding domain of its own and autoinhibited constitutively. However, HER2 forms a heterodimer with other ligand-bound EGF receptor family members, therefore stabilizes ligand binding and enhances kinase-mediated activation of downstream molecules. HER2 plays a key role in development, cell proliferation and differentiation. HER2 gene has been reported to associate with malignancy and a poor prognosis in numerous carcinomas, including breast, prostate, ovarian, lung cancers and so on.

Guidelines

2e5 of transient transfected anti- Her2 ScFv CAR-293 cells were stained with 1ug FITC-Labeled Her2 Fc Chimera Protein, Human, (Cat. No. UA011371) and unlable respectively (Fig. C and B), and non-transfected 293 cells were used as a control (Fig. A). FITC signal was used to evaluate the binding activity.
2e5 of transient transfected anti- Her2 ScFv CAR-293 cells were stained with isotype and Whitlow/218 Linker-Alexa Fluor® 488 (Fig. D and E). Alexa Fluor® 488 signal was used to evaluate the binding activity.
