Background

Fibroblast Growth Factor Receptor 1 (FGFR1) is a transmembrane receptor tyrosine kinase encoded by the FGFR1 gene that plays a pivotal role in regulating embryonic development, tissue repair, angiogenesis, and metabolic homeostasis by binding to fibroblast growth factors (FGFs) in conjunction with heparan sulfate proteoglycan co-receptors. Upon ligand binding, FGFR1 dimerizes and undergoes autophosphorylation on specific tyrosine residues within its intracellular kinase domain, thereby activating critical downstream signaling cascades including the RAS-MAPK, PI3K-AKT, PLCγ, and STAT pathways that govern cell proliferation, differentiation, migration, and survival. The protein's functional versatility is further enhanced by alternative splicing of exons encoding the third immunoglobulin-like domain (IIIb and IIIc isoforms), which dictates ligand-binding specificity and tissue-specific expression patterns essential for proper organogenesis and skeletal formation. Clinically, dysregulation of FGFR1 through gain-of-function mutations, chromosomal translocations such as t(8;22) in myeloid/lymphoid neoplasms, or gene amplification is implicated in various pathologies including craniosynostosis syndromes (e.g., Pfeiffer syndrome), osteoglophonic dysplasia, hypogonadotropic hypogonadism, and multiple cancers such as squamous cell lung carcinoma, breast cancer, and glioblastoma, making it an important therapeutic target for selective tyrosine kinase inhibitors and monoclonal antibodies currently under investigation in precision oncology.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human kidney. Anti-FGF receptor 1 antibody was used at 1/500 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Anti-FGF receptor 1 antibody was used at 1/500 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in A204 cells (top panel) and negative staining in HT-29 cells (below panel). Anti-FGF Receptor 1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibod at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
