Background
Fibroblast growth factor 6 (FGF-6), also designated as HST2 or HBGF-6, is a secreted peptide growth factor belonging to the paracrine FGF subfamily. The FGF6 gene, located on human chromosome 12p13, encodes an 18.9 kDa protein that exhibits the highest expression in skeletal muscle and myogenic progenitor cells throughout embryonic development and adulthood. FGF-6 binds preferentially to fibroblast growth factor receptor 4 (FGFR4) with heparan sulfate proteoglycan co-receptor dependency, triggering downstream signaling through the MAPK/ERK1/2 and PI3K/AKT pathways.Functionally, FGF-6 operates as a dose-dependent regulator of myogenesis, promoting satellite cell proliferation at high concentrations and initiating myogenic differentiation via calcineurin-dependent pathways at lower concentrations. Beyond muscle physiology, FGF-6 exhibits pleiotropic functions in bone remodeling, iron homeostasis, thermogenesis, and oncogenesis, harboring both regenerative and tumorigenic activities.
