Background

FBXL12 (F-box and leucine-rich repeat protein 12, also known as FBL12) is a substrate-recognition subunit of SCF-type E3 ubiquitin ligases; through its F-box domain it binds the SKP1–CUL1 complex and recruits specific substrates for polyubiquitination and proteasomal degradation. Its better-characterized substrates include calcium/calmodulin-dependent protein kinase I (CAMK1) and members of the ALDH3 family: FBXL12 mediates CAMK1 degradation, thereby interfering with cyclin D1/CDK4 complex assembly, causing G1 arrest in lung epithelial cells, and regulating T cell differentiation in a cell-autonomous manner. During placental development, FBXL12 downregulates ALDH3 activity by mediating its ubiquitination and degradation, thereby initiating the differentiation program of trophoblast stem cells; Fbxl12-deficient mice die during embryogenesis or perinatally due to impaired formation of the placental junctional zone. In addition, recent studies have revealed that FBXL12 also targets CHK1-phosphorylated FANCD2 for proteasomal degradation, thereby regulating replication fork restart and the replication stress response, and this FBXL12–FANCD2 axis is often overexpressed in highly proliferative tumors. Mouse genetic data show that Fbxl12 knockout leads to partial postnatal lethality, growth retardation, and abnormal placental vascular morphology.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human kidney. Anti-FBXL12 antibody was used at 1/400 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in HEK293 cells. Anti-FBXL12 antibody was used at 1/2000 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
