IHC shows positive staining in paraffin-embedded mouse testis. Anti-ETD antibody was used at 1/2000 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Background
Embryonic Testis Differentiation Protein (ETDP), often referred to in genomic databases as Embryonic Testis Differentiation protein homolog C (ETDC), is a protein-coding gene product primarily identified through homology in mammals (including humans and mice) that plays a specialized role during early male sexual development. Although its precise molecular mechanism remains an active area of research, ETDC is characterized by its specific expression in the embryonic testes, where it is hypothesized to contribute to the differentiation and maintenance of somatic cell lineages (such as Sertoli or Leydig cell precursors) or the regulation of germ cell quiescence following the sex-determination switch initiated by the Sry gene. As a homologous protein, it likely functions within the complex signaling network that ensures the proper morphogenesis of the testis cord and the suppression of female developmental pathways in XY embryos, making it a potential marker for studying disorders of sex development (DSD) and understanding the fine-tuned genetic control of gonadal differentiation before birth.
