Background

Extracellular signal-regulated kinase 5 (ERK5), also known as MAPK7 or Big MAP Kinase 1 (BMK1), is a unique member of the mitogen-activated protein kinase (MAPK) family distinguished by its unusually large C-terminal tail that contains a transcriptional activation domain and a nuclear localization signal, enabling it to function as both a kinase and a direct transcriptional regulator. Activated specifically by the upstream kinase MEK5 in response to growth factors, cytokines, and various stress stimuli such as oxidative stress and fluid shear stress, ERK5 plays critical roles in regulating cell survival, proliferation, differentiation, and angiogenesis through the phosphorylation of cytoplasmic substrates like MEF2 transcription factors and RSKs, as well as through direct gene regulation upon nuclear translocation. Its distinct signaling properties and essential functions in cardiovascular development, neuronal survival, immune responses, and tumorigenesis have positioned ERK5 as a significant therapeutic target, particularly in cancers where its overexpression correlates with poor prognosis and metastasis, although drug development remains challenging due to the need for high selectivity against other MAPK family members.
