Background

EIF4A3 (eukaryotic initiation factor 4A3) is a member of the DEAD-box RNA helicase family and an ATP-dependent RNA helicase that serves as the core scaffold protein of the exon junction complex (EJC). It adopts a butterfly-shaped conformation through its N-terminal and C-terminal RecA-like domains, which are used to tightly assemble with other core EJC components (such as Y14, Magoh, and MLN51) into a stable heterotetrameric core complex. The EJC is deposited approximately 20–24 nucleotides upstream of mRNA exon-exon junctions during splicing, where EIF4A3 binds and locks specific structures on spliced mRNAs to ensure stable assembly, thereby playing crucial roles in multiple post-transcriptional regulatory processes, including mRNA export, nonsense-mediated mRNA decay (NMD), translation efficiency, and localization. Dysfunction of EIF4A3 is associated with its aberrant expression in various cancers: in hepatocellular carcinoma, high expression of EIF4A3 promotes cell proliferation and metastasis; in glioblastoma, upregulated EIF4A3 expression is also correlated with poor patient prognosis and can influence the tumor microenvironment and immune responses through regulation of NMD pathways of target genes. Furthermore, loss-of-function mutations in EIF4A3 cause a rare autosomal recessive disorder known as Richieri-Costa-Pereira syndrome, which is primarily characterized by severe distal limb malformations, cleft palate, and intellectual developmental delay.
