Background

eIF3G, also known as eukaryotic translation initiation factor 3 subunit G or p35, is a crucial component of the eIF3 multi-protein complex, which serves as the largest and most complex initiation factor in eukaryotic translation. Structurally, eIF3G contains multiple RNA recognition motifs (RRMs) that enable it to directly bind specific mRNA sequences and interact with other eIF3 subunits, thereby playing a pivotal role in stabilizing the eIF3 complex and facilitating the assembly of the 43S pre-initiation complex at the 5' cap of messenger RNAs. Beyond its canonical function in promoting the recruitment of the 40S ribosomal subunit to mRNA, eIF3G has been implicated in the selective translation of specific transcripts involved in cell cycle regulation and stress responses, and its dysregulation or overexpression has been frequently observed in various human cancers, suggesting its potential as a biomarker and therapeutic target in oncology due to its influence on protein synthesis rates and cellular proliferation.

Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
