Background

EIF2S2 (eukaryotic translation initiation factor 2 subunit 2, also known as eIF2-β) is the β subunit of the heterotrimeric eIF2 complex, composed of 333 amino acids with a molecular weight of approximately 38 kDa, and it plays a key role in the initiation phase of protein synthesis. The eIF2 complex forms a ternary complex with GTP and initiator tRNA, binds the 40S ribosomal subunit, and subsequently recruits mRNA to assemble the 43S preinitiation complex. Structurally, EIF2S2 contains a highly charged N-terminal region (harboring multiple polylysine blocks), a zinc finger motif, and a C-terminal eIF2-β/eIF-5 homology domain, features that together confer mRNA-binding capability. Functionally, eIF2-β participates in regulating the activity of the eIF2 complex and influences the accuracy of translation initiation and the recycling of eIF2 through interactions with factors such as eIF5. Recent studies have found that EIF2S2 is highly expressed in a variety of tumors, including breast cancer, hepatocellular carcinoma, and glioblastoma, and is associated with tumor aggressiveness and poor prognosis; by regulating selective translation, it affects the expression of downstream genes such as ATF4 and CHOP, thereby participating in the regulation of cellular stress responses, proliferation, and apoptosis.
