Background

eEF2K (eukaryotic elongation factor 2 kinase) is a highly conserved Ca²⁺/calmodulin-dependent serine/threonine kinase that acts as a critical metabolic checkpoint by phosphorylating and inactivating eukaryotic elongation factor 2 (eEF2) at Thr56, thereby shutting down the energy-intensive translocation step of ribosomal translation elongation to conserve ATP during nutrient starvation, hypoxia, or oxidative stress; beyond this canonical translational control, eEF2K also exerts non-canonical, kinase- or eEF2-independent functions—modulating transcription factors such as p53, promoting DNA-damage repair, maintaining mitochondrial quality, and enhancing tumor cell survival in nutrient-poor microenvironments—while its own activity is tightly regulated by multiple upstream signaling pathways including mTORC1-mediated inhibition and AMPK- or TRPM7-mediated activation, making it a potential biomarker and therapeutic target in cancer and metabolic diseases.

Immunocytochemistry

ICC shows positive staining in HeLa cells. Anti- eEF2K antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
