Background

DOCK11, also known as Zizimin2, is a guanine nucleotide exchange factor (GEF) that plays a particularly important role in the immune system. Its core function is to serve as a specific activator of the small GTPase Cdc42, promoting the dissociation of GDP from Cdc42 and the binding of GTP, thereby regulating the formation of filopodia and cell migration within the cytoskeleton. DOCK11 is abundantly expressed in hematopoietic tissues such as bone marrow and lymphoid tissues, and is critical for B-cell development, secondary antibody responses, and the function of regulatory T cells (Tregs). Loss of its function leads to impaired Cdc42 activity and aberrant STAT5 signaling, and is associated with an X-linked early-onset autoimmune disease, characterized by immune dysregulation, cytopenia, and systemic lupus erythematosus-like symptoms in patients. Furthermore, DOCK11 has also been implicated in sustaining hepatitis B virus in hepatocytes and has been found to localize in the nucleus, suggesting potential unknown nuclear functions.
