Background
DOCK11, also known as Zizimin2, is an atypical guanine nucleotide exchange factor (GEF) encoded by the DOCK11 gene. It belongs to class D of the DOCK protein family and possesses a unique structure that lacks the conventional Dbl homology (DH) domain; instead, it exerts its catalytic function through its C-terminal DHR-2 domain, specifically activating the small GTPase CDC42. Functionally, DOCK11 is predominantly expressed in hematopoietic cells and lymphocytes, where it regulates dynamic actin cytoskeleton reorganization by activating CDC42, thereby promoting filopodia formation and influencing lymphocyte polarization and migration. This function is critically important in the immune system; studies have confirmed that loss-of-function mutations in the DOCK11 gene cause an X-linked early-onset multisystem autoinflammatory disease characterized by immune dysregulation, autoimmune cytopenias, and systemic lupus erythematosus. Lymphocytes from patients with this disease exhibit reduced CDC42 activity, impaired migration, and excessive production of pro-inflammatory cytokines due to DOCK11 deficiency.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human testis. Anti-DOCK11 antibody was used at 1/500 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
