Background

DNMT3A (DNA methyltransferase 3 alpha) is a crucial de novo DNA methyltransferase enzyme in mammals that establishes new cytosine methylation patterns, primarily at CpG dinucleotides, during early embryonic development and germ cell formation, distinct from DNMT1 which maintains existing methylation marks. Structurally, it contains a catalytic C-terminal domain and an N-terminal regulatory region with ADD and PWWP domains that facilitate interaction with histone tails and chromatin, thereby guiding precise genomic targeting. Beyond its canonical role in epigenetic silencing and regulation of gene expression, DNMT3A is vital for cellular differentiation, genomic imprinting, and X-chromosome inactivation; notably, somatic mutations in the DNMT3A gene, particularly at residue R882, are frequently observed in acute myeloid leukemia (AML) and other hematologic malignancies, as well as in overgrowth syndromes like Tatton-Brown-Rahman syndrome, highlighting its dual significance as a key regulator of normal development and a critical tumor suppressor whose dysfunction leads to aberrant hypomethylation and oncogenic transformation.
