Background

DLX3 (Distal-less 3 homeobox protein) is a homeodomain-containing transcription factor belonging to the DLX family, playing critical regulatory roles in embryonic development, skeletal formation, and tissue differentiation. The core structural feature of this protein is its highly conserved homeodomain, which specifically recognizes and binds to DNA sequences in the promoter regions of downstream target genes, thereby activating or repressing gene transcription. The expression pattern of DLX3 exhibits significant tissue specificity, being predominantly enriched in the placenta, tooth follicles, and bone tissue, where it finely regulates the differentiation and maturation of osteoblasts, odontoblasts, and trophoblast cells through synergistic interactions with signaling pathways such as BMP and WNT. Clinical and genetic studies have shown that gain-of-function or loss-of-function mutations in the DLX3 gene can lead to a spectrum of autosomal dominant disorders, including tricho-dento-osseous syndrome and amelogenesis imperfecta with taurodontism, which are primarily characterized by enamel mineralization defects, skeletal abnormalities, and sparse curly hair. Furthermore, DLX3 also plays important roles in osteoclastogenesis and epidermal barrier formation, and its dysregulation may be associated with the development of osteoporosis and certain skin diseases, underscoring the research value of this transcription factor in developmental biology and regenerative medicine.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human tonsil. Anti-DLX3 antibody was used at 1/2000 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in JAR cells. Anti- DLX3 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
