Background
Desmoglein-3 (Dsg3) is a calcium-dependent transmembrane glycoprotein and a key component of desmosomes, specialized cell-cell adhesion structures primarily found in stratified squamous epithelia such as the skin and oral mucosa; it functions to maintain tissue integrity by anchoring intermediate filaments of the cytoskeleton to sites of intercellular contact, and beyond its structural role, Dsg3 actively regulates epithelial homeostasis by suppressing p53-mediated stress responses, thereby acting as an anti-stress protein that protects keratinocytes from apoptosis induced by UVB, mechanical stress, or genotoxic agents, while also promoting cancer cell migration and invasion through activation of the AP-1 transcription factor and PKC-dependent phosphorylation of ezrin, and its autoimmune targeting by autoantibodies results in the loss of adhesion and blister formation characteristic of pemphigus vulgaris, making Dsg3 a central player in both physiological epithelial cohesion and pathological processes including cancer metastasis and autoimmune blistering diseases.
