Background
DDX54, also known as DP97, is an ATP-dependent RNA helicase encoded by the DDX54 gene and belongs to the conserved DEAD-box protein family (named after the characteristic Asp-Glu-Ala-Asp amino acid motif). This protein is primarily localized in the nucleolus and plays indispensable roles in a variety of biological processes, including ribosome biogenesis, precursor mRNA splicing, transcriptional regulation, and DNA damage response. At the molecular level, DDX54 can unwind RNA duplex structures in an ATP-hydrolysis-dependent manner. Its functional specificity is reflected on one hand in its hormone-dependent binding to nuclear receptors, acting as a transcriptional corepressor that inhibits their transcriptional activity; on the other hand, upon genotoxic stress such as ionizing radiation, it responds immediately and binds to a class of precursor mRNAs containing weak splice acceptor sites, modulating transcriptomic dynamics by enhancing splicing efficiency, thereby promoting cell survival. Furthermore, DDX54 is indispensable for central nervous system myelination, and its functional aberrations or overexpression are closely associated with the development and poor prognosis of various cancers, such as colorectal cancer and hepatocellular carcinoma, making it a potential drug target or prognostic biomarker.

Immunocytochemistry
ICC shows positive staining in HeLa cells. Anti-DDX54 antibody was used at 1/50 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
